The detection of the main causes of plant disease, pathogens in particular, is a prerequisite for diagnosing a particular disease of plants. The diagnosis must meet a standard procedure known as Koch’s postulates. The details are; 1) there must be a primary pathogen in diseased plant parts, 2) it must be isolated and cultured in pure culture, 3) if the pathogen from pure culture was inoculated into the same host plants, make the plant sick with similar symptoms, and 4 ) pathogen must be isolated and cultured in the artificial medium. It can be seen that the artificial medium is an important aspect that must be held prior to study further.
The problem that arises is when the ingredients for growing pathogens or pathogen growing medium are difficult to find or hard to identify so that researchers difficult to determine the proper application of the growing medium for research. For example, for diseases caused by pathogens, such as rust caused by Puccinia spp., Flour, and so on, which still has not found a suitable artificial medium for growth. In addition, the undiscovered growing medium will lead to the issue of specimens originality that were used as the parent object that should be maintained. In a study using microbes, laboratory testing is need to be done first before being applied in the field scale. In testing the effectiveness and dosage of certain pesticides, it is also required laboratory test using a medium cup.
Those are the things that important in research of artificial medium. This book answers these problems. Ika Rochdjatun expound on recipes to make different types of artificial medium that can be used for research in plant histology laboratories, among others: artificial medium for fungi, artificial medium for bacteria, and artificial medium for histology test plants. Therefore, the reader of this book will be able to know in detail about how the procedures or techniques to create an artificial medium, supplemented with the right composition to make it. No lag, the authors also added discussion about observation techniques under the microscope, accompanied by pictures on their observations.
Ika also systematically review on the different types of existing medium, such as a simple medium, medium natural, enriched medium, minimal medium, selective medium, and so forth. Any knowledge of the artificial medium is certainly very helpful for readers who want to test the suitability of the medium for a certain microbes or pathogens. Diagnosa suatu penyakit pada tumbuhan memfokuskan diri pada patogen sebagai penyebab utama penyakit tumbuhan. Postulat Koch menyebutkan empat rincian prosedur standar dalam diagnosa penyakit tumbuhan, diantaranya; 1) patogen penyebab utamanya harus terdapat pada bagian tanaman sakit, 2) harus dapat diisolasi dan dibiakkan dalam biakan murni, 3) apabila patogen dari biakan murni itu diinokulasikan ke tanaman inang yang sama maka harus membuat tanaman tersebut sakit dengan gejala yang serupa, dan 4) patogen harus dapat diisolasi kembali dan dibiakan dalam medium buatan.
Permasalahan yang kemudian timbul adalah ketika bahan untuk menumbuhkan patogen atau medium tumbuhnya sulit dijumpai atau sulit diketahui. Buku ini hadir untuk menjawab persoalan tersebut.
